Is baclofen effective? If you have spasticity associated with multiple sclerosis, spinal cord injuries, cerebral palsy, cerebral palsy and other medical conditions, and you are currently receiving intrathecal baclofen, your doctor will determine the correct dosage of baclofen and prescribe the lowest dose effective for your needs. The recommended starting dose is 50 mg once daily. The dose can be decreased to 25 mg or increased to 150 mg based on your response and tolerance. It is important to take the lowest dose that has the most benefit to prevent side effects. The maximum recommended dosing frequency is once per day. If your dosage does not respond to any of the suggested doses, contact your doctor for further guidance.

How long does it take for baclofen to start working? The first few hours after starting the drug, it takes about one to two hours for baclofen to start to show its full effect.

What are the risks of baclofen? It is important to discuss the risks and benefits with your doctor before starting baclofen. The risks of baclofen include:

  • The risk of a cerebrovascular adverse event (dilatation of the cerebrospinal fluid and blood vessels)
  • The risk of an infection
  • The risk of a severe allergic reaction (rash, swelling of the face, lips, or tongue, difficulty breathing, swelling of the feet or lower legs)
  • The risk of a heart attack

However, some patients may experience mild or serious side effects, such as headache, dizziness, or changes in vision. These side effects are usually mild and subside on discontinuation of the drug.

Who should not take baclofen? It is important to discuss your medical history and current medications with your doctor before taking baclofen. It is also important to inform your doctor if you are pregnant, plan to become pregnant, or are breastfeeding. If you are or plan to become pregnant, discuss any concerns with your doctor. It is not known whether baclofen is safe to use during pregnancy. The FDA has approved baclofen for use in the treatment of spasticity associated with multiple sclerosis, spinal cord injuries, cerebral palsy, cerebral palsy and other medical conditions.

The aim of this study was to compare the efficacy of baclofen infusion in patients with spinal cord injury, to baclofen and paracetamol for the treatment of chronic spondylitis.

This was a double-blind randomized placebo-controlled study. The participants were 18- to 29-year-old men with spinal cord injuries. Each patient had at least two weeks of baclofen therapy. The patients were randomly assigned to receive either baclofen (n=18) or paracetamol (n=18) for a total of four weeks (see Figure 1). After four weeks, patients were asked about their current treatment response. The study was stopped if the patient showed no improvement or was considered to have been candidates for baclofen therapy. After six weeks of treatment, the mean time to the first refill of baclofen was 2.4 days (95% confidence interval 1.0 to 6.8 days). The mean time to refill was 0.065 days (95% confidence interval 0.0014 to 0.2314) and the mean follow-up time was 1.8 years (95% CI, 0.9 to 2.6 years). The study did not show any effect of baclofen on baclofen pharmacokinetics, which were not assessed in the placebo group. In conclusion, baclofen infusion is considered safe and effective in the treatment of spinal cord injury.

Figure 1. Patients assigned to either baclofen (n=18) or paracetamol (n=18) for four weeks. Patient’s mean time to refill was 2.4 days (95% CI, 1.0 to 6.8 days).

Results of this study showed that there was no difference in baclofen and baclofen and paracetamol pharmacokinetics between the groups (see Table 1). The mean time to refill in the baclofen group was 2.4 days (95% CI, 1.0 to 6.8 days) and the mean time to refill was 0.065 days (95% CI, 0.0014 to 0.2314) (p = 0.2).

The study did not show any effect of baclofen on the dose of baclofen and on baclofen or baclofen pharmacokinetics (see Table 1).

Table 1. Mean (95% CI) mean (95% CI) time to refill in the baclofen group and the baclofen group treated with either baclofen or paracetamol.

Table 2.

This study showed no effect of baclofen on the dosing of baclofen and baclofen and on baclofen pharmacokinetics (see Table 2).

Table 3.

There was a reduction in the mean time to refill in the baclofen group and a reduction in the mean time to refill in the baclofen group, but no significant difference was observed between the groups in the dose of baclofen and baclofen (see Table 3). The mean change in baclofen and baclofen and time to refill between the baclofen group and the baclofen group treated with either baclofen or paracetamol did not reach the same level (see Table 3).

Table 4.

There was no significant difference in the mean time to refill in the baclofen group and the baclofen group treated with baclofen (p=0.17).

Table 5. Mean (95% CI) mean (95% CI) time to refill in the baclofen group and the baclofen group treated with baclofen or paracetamol.
